Specific receptors as well as ectonucleotidases (NTPDases) mediate and control the biological action of extracellular nucleotides. The sensitivity to ATP of differentiating satellite and C2C12 cells was investigated. Proliferating myoblasts and myotubes expressed five P2X (1, 3, 4, 6 and 7) and four P2Y (1, 2, 4 and 6) receptors and four NTPDases (1, 2 and 3 and a-sarcoglycan). However, the expression level of receptors and NTPDases, thus responsiveness to ATP action, changed dramatically during differentiation. Ecto-ATPase activity of cells progressively increased in parallel with myotube formation. Mechanical or electrical stimulation of myotubes produced significant release of ATP. The nucleotide appears to stimulate activation, mobilization and proliferation of myoblasts, while the prolonged exposure to and excessive levels of ATP compromise the survival of myotubes.
