A polymeric drug delivery system for the release ofnarciclasine, a natural substance with high antimitotic and potential antitumor activity, was prepared by radiation-inducedpolymerization of methacrylic monomers. A wide range of narciclasinerelease rates was obtained with polymers based on 2hydroxyethyl methacrylate to which different amounts of a crosslinking agent or MPEG-350 were added. A study of the narciclasinerelease curves indicated that a unique mechanism of release does not hold for all matrices. The release of narciclasine from polymer matrices occurs ‘in vivo’ without the deadly effects observed when the same dose of drug is directly injected. The narciclasinerelease is accompanied by inhibition of growth of mice, as expected from the antimitotic effect of the substance.
