The activity of 3-carbethoxyangelicin (3-CA). an angelicin derivative carrying an electron withdrawing group at pyron side, has been studied in Saccharomyces cerevisiae D7. By UVA irradiation, 3-CA showed a reduced killing effect, similar to that provoked by 3-carbethoxypsoralen (3-CPs), but much lower than 8-methoxypsoralen (8-MOP), both used as references. Contrary to 3-CPs, 3-CA did not induce rnitochondrial (Rho-) mutants, a behaviour generally observed with rnonofunctional furocoumarins, while no double-strand breaks are observed after post-treatment incubation and separation of yeast chromosome by pulsed-field electrophoresis. These observations suggest 3-CA can kill yeast cells by a mechanism different from DNA photobinding typical of known furocoumarins, probably by an extensive induction of single-strand break in DNA.
